Note: The first two hours of Housefull 5B are identical to Housefull 5A; the difference lies entirely in the final reveal and the identity of the main conspirator.
The Climax of Housefull 5B (SPOILER)
The "B" version dramatically changes the villain to one of the corporate characters:

Mastermind: Maya (Chitrangada Singh), the company's cunning CFO, is revealed to be the main orchestrator of the crimes.

Motive: Maya was deeply in debt from gambling and planned the murders and the inheritance fraud to gain access to Ranjeet's vast fortune.

Accomplice/Second Killer: Jalbhushan / Jolly 2 (Abhishek Bachchan) remains her partner in crime (same as in Housefull 5A), carrying out the murders at her direction.

The Murders:

Maya killed the doctor after he caught her trying to tamper with the DNA test results.

Maya also killed Bedi when he discovered evidence.

Jalbhushan killed Captain Sameer as he was a witness to one of the crimes.

The Real Jolly: Similar to the other version, Bobby Deol makes a cameo appearance as the true heir, Jalal, who ultimately decides to share the inheritance with the main characters.
In summary, the key difference is that in 5A the mastermind is Ranjeet's son Dev (Fardeen Khan), while in 5B the mastermind is the CFO Maya (Chitrangada Singh). Abhishek Bachchan's character, Jalbhushan, is an accomplice in both endings.
